It has been reported that in normal cells axin1, APC, WTX, and β-catenin form a complex and that axin1 also interacts with many other proteins such as RanBP9, USP7, and so on. And USP7 interacts with ataxin-1, whose phosphorylated form is stabilized by 14-3-3. So I wish to raise the following two questions:

1 Can we determine the stoichiometry of full-length human axin1-APC-WTX-β-catenin-RanBP9 complex using AFM in solution?

2. Can we observe the conformational change of 14-3-3 induced by its binding to phosphorylated ataxin-1 using AFM in solution?
